eMolecules compound 6142342 was purchased from ChemDiv (San Diego, CA), initially dissolved in dimethyl sulfoxide, and then further diluted in assay buffer. For kinetic assays carried out as described for the steady-state kinetic assays described above, the final solution contained 320 pM purified MBL, 10 microg/ml BSA, and either no or 40 microM compound 6142342 in the kinetic assay buffer described above plus 2 mM 3-[(3-cholamidopropyl)-dimethylammonio]-1-propanesulfonate (CHAPS) detergent. The percent residual activity was defined as the initial velocity for the conversion of 6 microM chromacef in the presence of compound 6142342 divided by the initial velocity without the compound. Percent inhibition was defined as 100% minus the residual activity. The experiments were carried out in triplicate.
